A third digit 0 to 5 is a natural person's RUC (the cédula plus 001) and is refused, and so is a RUC ending 002 or higher, which is an establishment. A 12-digit value is read with the zero of the province restored (a spreadsheet drops it). The check digit is only a guide: the numbers of the Sociedades por Acciones Simplificadas are plain serials that often fail it. The RUC is the tax number: there is no separate VAT number.

A valid shape and check digit prove that a number is well-formed, not that a company holds it. Only a lookup in the register answers that.
